Shrines are statues that the player can interact with to (sometimes) sacrifice something and receive something in return. Shrines can be indicated before entering due to the presence of a green lantern by the door.

Standing near highlighted enemies damages and drains them, healing the player after draining enough. This can be used to damage enemies such as Cannonbalrog while they have disappeared. Costs 10 to use the first time, and the cost increases by 10 per use. Every time the player fires a weapon, there is a chance for it to quickly fire 2 to 4 times at no extra ammo cost. Each use linearly increases the chance to activate the effect by 3.7%.
Repeated use only increases the chance to fire additional bullets. It has no effect on the number of bullets fired — these are always random. |

- Prior to the Supply Drop Update, the Dice Shrine's Limited effect decreased maximum ammo by 50% instead of 30%.
- Prior to v1.1.3, the Dice Shrine's Unsteady effect decreased reload time instead of increasing it.
- The Dice shrine can apply two opposite effects at the same time, such as Renewed and Pained, Paid and Robbed, Cleansed and Cursed, or Blanked and De-Blanked.
- The Dice Shrine's rare effect will not reduce The Robot's armor when triggered.
- The Blood shrine's blessing can carry over to a character's past.
- Using the Y.V. Shrine 27 times, for a total cost of 3780 , gives every shot its bonus. However, obtaining this much is difficult. It requires lots of curse, money items like Coin Crown, and as much Gold Junk as possible. Alternately, the infinite money exploit with Blobulin Rancher and Don't Blink in Challenge Mode can be used to obtain this much money. Another possibility is through the use of Lil' Bomber in conjunction with its King Bomber synergy.
- The Y.V. Shrine's bonus does affect shots fired from companions; however, they will be shot in a straight line going to the right, instead of being aimed at enemies.
- The Peace Shrine does not accept weapons with infinite ammo like Casey.
- The Peace Shrine will accept a gun from The Robot, but will not return any benefits.
- During a Rainbow Run, The Companion Shrine will not give you a companion, but still will take one of your heart containers.
- The Junk Shrine will not take Lies, Gold Junk, or Ser Junkan.
Trivia[ | ]
- The Blood shrine subtitle "Kaliber Mah Shakti De" is a reference to "Kali Ma Shakti De", which is spoken by Mola Ram in the movie Indiana Jones and the Temple of Doom before he sacrifices a victim by pulling out his heart, thus the loss of a heart container upon using the shrine. "Kali Ma Shakti De" is a phrase in the language of Hindi, praying for strength from the Hindu goddess of Death and Time, Kali.
- The Y.V. shrine refers to Y.V. (Yung Venuz) from Vlambeer's Gun Godz and Nuclear Throne games. Y.V.'s active ability in Nuclear Throne, called 'Pop Pop', allows him to fire a weapon twice (or 4 times when the ability is upgraded) with one pull of the trigger. Needing to sacrifice coins to the shrine for his blessing (and Y.V. being depicted standing on stacks of paper bills) refers to his love of money, of which he has a great deal.
- In Nuclear Throne, Y.V. himself happens to be voiced by Enter the Gungeon's composer, doseone.
- The Hero shrine depicts the Lich or Gunslinger with his head missing.
- The Blank shrine depicts Old Red, who appears in-game as a random shopkeeper.
- The Companion shrine depicts the Super Space Turtle, Blank Companion's Ring, Baby Good Mimic, R2G2, Pig, Wolf, Dog, and Badge.
- The Companion shrine's subtitle refers to the phrase “A friend in need is a friend indeed”.
- Shell'tan is a reference to the villain Shai'tan in the Wheel of Time series, in addition to referring to the Muslim name for the Devil (Shayṭān).
- The pose being struck by the woman depicted in the Cleanse shrine is known as "Finger Guns", and a reference to the Buddy Christ statue from the 1999 religious comedy film Dogma.
- The internal variable for the Dice Shrine's chance to explode is called 'ChanceToGoApeshit'.
